Thats why i'm asking does my auto sensor cleaning is enough to work like this or not, because turning off the camera doesnt have the same aperture as like as we set.

I'm using Canon EFS kit lens, and planning to buy a 50mm Prime lens. Though my 600d have built-in sensor cleaning, can i reverse mount the lens after setting the aperture by straight mount with holding the DOF preview button? or What is the best way to set the aperture?
Yep, that's how you set aperture on an EF lens. A very clumsy work-around. You're probably better off buying a non-EF lens for this, like an AI Micro Nikkor. At least you'll be able to set the f/stop.

And I have no idea what this has to do with sensor cleaning.

Readed somewhere in an article that removing lens with camera on attracts dust twards the sensor.
it doesn't attract dust but don't leave the camera without a lens or cap

Using a reversed lens is inexpensive but it can be frustrating. (extremely narrow DOF)

Go ahead and get a $5 adapter, use the DOF preview button to change aperture and see if it's for you. If you do any macro shooting you're probably want to get a real 1:1 macro lens such as the Canon 60mm, 100mm or Tamron 90, Sigma 105, etc.

when you reverse the lens, you can't adjust any aperture. The lens electronic contact doesn't connect with the body, so on or off it doesnt matter for the setup. You can adjust aperture if you have lens that has manual aperture ring on the lens itself.
