Being serious for a second, to most native English speakers, it means "the best", or "perfect" choice. There's also often an implication that it doesn't actually exist. The original Holy Grail is part of the legend of King Arthur.

Generally what Holy Grail means is the ultimate, the best, the one everyone wishes for.
