By lock is the reflex mirror locking up?  If so, what happens inside the mirror box?  What is exposed, the shutter blades or the sensor itself?  Let's go over the most likely causes in each situation.  Assuming we are seeing the reflex mirror lockup, but the shutter blades stay down, then the most likely cause is the shutter assembly itself (it's a very complicated assembly and could be any one number of things).  If the reflex mirror stays locked down it's most likely the shutter block.  If the sensor is exposed, I would suspect those sensor flex cables or possibly the rear screen itself.  What does the camera do using the viewfinder?
