Macro Magnifcation Query

Hi all,

Using a Canon R6 and wanting to learn more with macro photography for insects / small critters.

I have a Canon 100 f/2.8 USM macro lens (non-L) and Canon 100-400 IS II lens (with 1.4x Extender)

I also have the Kenko Extensions tubes, and when stacked, amounts to 68mm in total

And finally the Raynox DCR-250.

Could someone kindly work out the magnifcation achieved with the following combinations and the closest working distance please?

Canon 100mm macro with Raynox DCR-250?

Canon 100mm macro with Raynox DCR-250 AND extensions tubes stacked?

Canon 100-400mm IS II with Raynox DCR-250 (presumably using the 40omm end) (would I focus to infinity to achieve largest mag?)

Canon 100-400 IS II with 1.4x Extender and Raynox DCR-250?

Finally, with the R6 the internal focus stacking workings olny if the autofocus is left on, but presumingly I would be focused to 1:1 with the Canon macro lens.

So I presume just focus stack by moving back and forth, overlapping each frame?

Do you use manual mode, or Aperture mode?

FInally do you use flash (and if so on TTL or Manual and which softbox/diffuser would you recommend)? (Ideally I want to use ambient / natural light) or do you just bump up the ISO?
