You say you are in "manual". You mean you have the mode dial set to M on top? So you are setting the exposure and shutter speed manually? What about the ISO? If your ISO is fixed, the exposure compensation dial will have nothing to do. It only works to modify exposure when at least one of the three variables (shutter speed, aperture, ISO) is set to auto.
