What does DO mean and why do we need it?

DO stands for Diffractive optics. It is a Canon propriatry system.

Nikon's equivalent system is called PF short for Phase fresnel.

Both systems use some form of fresnel lens to bend the light more than a heavier glass lens, hence the shorter length.

Nikon's 400mm f4PF lens is a miniature marvel for APSC or full frame, but has strange bokeh effects in some situations.
