I think, the Fuji 100-400mm lens is currently the best 400mm telezoom that is available for mirrorless, especially because it can be combined with the 1.4x teleconverter, and this converter is not too expensive and can be purchased used.

So manual focusing is also an option for me and the fuji lenses support this very good, so far I have seen on youtube (I did a lot of recherche)

There is one additional question: Is it possible to prefocus manually and afterwards use the (backbutton) AF for quick fine focus? If possible, this should increase the overall focus speed.

You can use MF+AF at the same time. Haven't tried it, but it should help.

In a Fuji camera with the focus mode selector in MF, you can assign AF-C to a function button and have continuos AF all the time. So you can manually pre-focus and then press the function button.
