Not only the aperture, but the focal range as well. I'm not going on safari, but the 12-40 works for 85% of the landscape I do. I often carry just that lens for that reason. On some trips I add the 35-100 F2.8 and the 8mm FE. The choice is based on the particular mountain trip and topography. Of course, with Safari, 100mm may not be enough. I carry and use the 35-100 for particular mountain trips and when I expect it likely that I will see large fauna like Mountain goats, Bighorn Sheep, and Bears.

The 12-100 is not too heavy for me to want to carry, but too large (as would be the 100-400) - but a safari would be different on the long end.

You should then be able to make an easier decision regarding the 12-100 F4 Pro
