Back button focus and metering?

I have always shot my photos using back button focus and then the shutter button to take a photo.

With spot metering, it says that you need to focus on a midtone, get an exposure then adjust, such as +1/2-2/3 of a stop for Caucasian people.

Does this mean every time I push the focus for a new shot now, before taking a photo to get the eyes in focus, that I’ve lost my exposure reading? I’m thinking so, so how would I lock the exposure in?

I use a 5d mark 3, and would appreciate any advice. Do I also need to use the middle focusing point on the 5d for spot metering? So lost :/

Thanks
