The 50mm f2.5 is an older FF lens, which only reaches 1:1 macro with its optional (and I am not sure if it is still widely available) 1:1 adapter (which is a mix of extension tube and tele converter).

The 60mm f2.8 from Canon is a good lens, also usable as portrait lens. However, personally I'd also look at the Tamron 60mm f2 Di II macro. It has a stop wider max. aperture, nice for when you use it for portraits, and it has a smoother bokeh (makes things not in focus have a smoother blur) than the Canon.

The Canon's plus over the Tamron then is the USM motor/AF.
