There is an excellent channel for Google Nik on YouTube.

The Sharpener Pro 3 series of Videos is a good resource. From memory, one of them guides through Raw Pre-Sharpening and subsequent output sharpening uses a picture of an alligator that I thought was particularly excellent and demonstrated things beautifully.

For my purposes I default in Lightroom to turn off the standard 25% sharpening. I run my images through Nik Sharpener Pre-Raw first. After my other edits (hopefully enhancements ),  I then localise my sharpening efforts in creative mode (i.e: no output destination) and save back again. At this point I only sharpen for output during export phase from Lightroom.

(My Photoshop procedure is similar, but I tend to do a lot more with masked Nik layers when going that route. Also smart layers make things much easier in many cases).
