Yes, it is possible to assign the "Shortcut" button as a back-button focus lock for the Canon G12. The Shortcut button is located on the back of the camera at the upper left corner. The graphic on the shortcut button is an "S" surrounded by a box with a small arrow through it.

To assign the Shortcut button for AF Lock:

Menu > Camera Tab > Set Shortcut button > Select AFL - AF Lock then press the Func Set button in the middle of the back dial.

To use, press the shortcut button to lock focus then compose and shoot.

I recommend setting the AF Frame to FlexiZone as well. This is found in the Camera Tab of the Menu also.
