Probably not. The Airport Express is a wireless access point / router. That is, it connects wireless clients such as WiFi-equipped Macs and PCs to each other and to a broadband network.

When you are traveling, you're going to need to connect to a hotel's wireless access point / router as a client, not as another access point. I don't know if the AirPort Express has a client mode; since that would rarely be useful for its primary purpose, I'd venture to say that it does not.

If your Mac does not have built-in Wifi, you can use an Airport Express to connect to a wireless network and share this connection via Ethernet to the Mac. See this article for the basic instructions.
