I think you mean 'how would one estimate or derive e1?', which is what one wants, and not estimate the MSE of e1. The MSE of e1 can be derived analytically, under the conditions stated in previous messages, and is equal to average number of photons impinged on a pixel times (1 - QE). Estimating e1 is again possible analytically, however, in practise can turn out to be difficult as it requires certain knowledge beyond just the number of photoelectrons given. Since many times the number of photoelectrons is all one has, going with e2 is perhaps the only choice.
