I like to use the Program mode and set it for spot focus, and sometimes use spot exposure if needed (great as long as I don't forget to put it back to the averaged exposure afterwards), or adjust the EV up or down as needed. You can pick your ISO, too.

The canned scene modes are also very good, if you don't mind the camera selecting the focus point--however, as a workaround, you can move the camera around and re-focus until it finally picks the focus point you want, and then recompose the picture. It does a beautiful job with the Landscape scene mode.
