custom white balance using digital calibration target

Good morning,

This is my first posting and if someone could provide the answer to my question it would be most appreciated.

I have been using Photovision Digital Calibration Targets in my studios for the past six months and have found them to be very effective in setting both aperature and white balance. However, I still am unconvinced that I fully understand what I am doing. First I fill the frame with the target. Sometimes in so doing I use my zoom. I am than able to get a reasonably good histogram with a good center blip.

My exposure is than set as is the white balance. If I than change my iso, aperature, shutter speed or focal length is my white balance going to be effected. I am spending too much time in PP and believe that I may not be using the target properly.
