Help! Advice / tips on the 100-400

I've recently procured a 100-400 with a 40d (moved from Olympus). My initial playing around with the lens gives me much joy but I feel that this lens needs more using / learning before I start seeing real results. I'm off to a tiger safari for a couple of days end of the month and do not wish to lose opportunities because of my limited experience with the new system. Is there any advice/tips you can share with me to optimize performance and to cut down the learning curve. All advice greatly appreciated.
