Dirt on D7000

Hi all,

When I point my D7000 at a bright object, eg. at the sky, there is a noticeable bit of dust/dirt. I have wiped the lens on both ends, as well as the viewfinder, and nothing's changed, so I assume this thing is inside the camera. I have tried running the sensor-cleaning menu option, to no effect. I know there's a mirror-up option on the D7000 that presumably allows me to clean inside, but am after advice... what do I need to do to get rid of this piece of dust? Mirror up and just wipe inside the camera with a clean cloth, or... ?

Thanks.
