Best techniques to scan a film negative

I recently bought a Epson v500 and have been scanning my 35mm film negatives in it. I set it to professional mode and either scan it at 4000 or 6400 DPI. My Jpeg's still come out around 5-6mb and not the 24mb size that everyone seems to be getting. If i scan Tiff files they are around 300mb. I use the epson software to scan.

I also noticed something weird. If i just import it to Lightroom and export without doing any processing at all, the files come out around 25 mb. Not sure why?

Can anyone share what are the best settings and techniques to scan the negatives using this scanner. Also shed some light on post processing images after they are scanned. Like does it even makes sense to process film images.
