My advice is take some time reading dpreview's article on how to set it up, then fiddle with different settings to see how you like them.  Try assigning different functions to different buttons.  Make the focus point tiny (I love this feature).  When you get comfortable with most of the features play around with the Art Filters; I thought they might be kind of cheesy, but find several of them most interesting.  I particularly like Key Line for people and animals, and Dramatic Tone for certain landscapes.
