Doubling the Ram to 8GB is a no brainer. You will notice that immediately. Moving the drive into the Mac Mini is also going to be a lot faster than the USB port. I would do both. You wont think it's the same machine.

The next real speed upgrade would be to replace the system drive with an SSD. They have come down in price now so it's something you might want to consider.
