I need the angle of view that a 35mm gives at about 0.3-0.5m distance. 50mm will be a bit on the narrow side, but I can move a bit further away if this gives a better focus field. I can't go much further than 0.5m though, so 50mm is stretching it. So, thus this trade-off.
Why f1.4? Any reason to think of such a large aperture? Diffraction softening should only set in past f4/5.6...

What kind of scientific project is it? MF will give better focus consistency.
I will need AF, the distance will alter from sample to sample slightly, and I'm looking at sub-millimeter DOF's.
The Nikon D800 has good AF, but not sub millimeter good at f1.4. The Canon 5D mk III has a bit more accurate AF, but you still might not get the performance you are thinking of... But that camera (obviously) has 10% less pixel width/height.
For this project, I will be taking pictures of low contrast objects at near distance, probably 0.3-0.5m. I need the "entire" frame to be captured in "perfect" focus left-right (top-down less important).
Focal length determines the distance. So again, why 35mm? Unless that has a real reason, I would think that you'd even be better served with a 1960's Nikkor 55mm f3.5 micro, pre-Ai, MF lens.
f3.5 is too small, but thank's for the suggestion.
As far as I know, no one tests (formally) for field curvature, so you will not get any wiser asking on here. I suggest you pay a visit to a shop in your vicinity, and quickly try a quick test with a FF camera body and your own card at f1.4.
