I use Fedora and their liveusb-creator can create live usb with persistence meaning it will save your changes upon reboot. Live usb's have a limited life across many saves though and eventually run out of space. In Fedora I have installed from DVD to usb and run it as a regular operating system the same as a hard drive install.
I think that Ubuntu may have "persistence" and I know that Knoppix has for sure with their live cd/dvd.

Yes, Ubuntu LiveCD have the "persistence" option. I recently played with a 2GB flash drive and that was sufficient for 500MB "persistence" drive on it.
