I tried your test setup and did see a small difference on the edges of the frames. (I made my own focus chart using a draw program). I used my 50 mm f1.4. My biggest issue is precisely placing the af point with the camera on the tripod... couldn't get the af point centered on the line exactly(using the viewfinder). So I would assume under extreme magnification there would be a difference on the edges of the frame.

All in all, I'm going to have to say the focus on my T4i is on..whether it is spot on is up for debate, but I don't think any mis-focusing issues would be readily seen.
