You will lose Dynamic range and low ISO. For measured values, check the camera ratings at: http://www.dxomark.com/
I used to own both cameras, but used the D300 mostly for sports, when the fps was important. For other photography, I mostly used the D5000, which not only has a better sensor but is also lighter and has a useful swivel screen.
That said, of course the build quality of the D300 is magnificent compared to the D5000, and it is a pleasure to hold it in one's hand. A good feel, does not rescue blown out highlights, however.
