I had D7000 and programmed the AE-lock button as AF-ON. The problem was that AF-ON was too close to the eye piece and so my thumb interfered with the spectacles.

The D600 layout is the same as D7000, but as the D600 is 9mm wider and from the pictures there appears to be a little more distance between eyepiece and AE-lock button. Hopefully it would be OK.
