Another position to check, is the Custom Functions in the Flash. You can access this either on the Flash, or through the camera itself.

Select External Speedlite Control (Shoot Menu 3) > Flash C.Fn setting > Option 8 needs to be set to 0. (8:0). This enables the AF-assist beam firing.

Nibby, thanks a lot, I had set my 1Dx at "Servo AF", that's why the AF assist beam did not work, once change to "one shot" AF, then the AF Assist Beam come, even with the old ST-E2. I totally cannot remember this restriction.
