Attach your 400mm to your camera and mount on a heavy tripod at the lens collar. Focus on something 50 feet away, turn on liveview and magnify to the maximum. Now touch the tripod or blow puffs of air at your camera and watch the image in liveview.

OK. Now I understand what you were saying.

My 5D3 just came in this afternoon and tomorrow I expect to begin checking to see how my glass does with it in a serious way. I have already tried some initial hand-held tests and it appears that my 100-400 is stop-on, my 70-300 DO appears to be a bit off (and is a bit soft under the best of settings) and the 24-105 that came with the camera kit seems to be just fine. I thought I would first check using the tips from this link - http://www.birdphotographers.net/forums/showthread.php/58042-AF-microadjustment-tricks - and then order the FoCal software and see how the two methods compare.

Thanks for your help.
