It is better to do all your editing in 16 bit. Before CS5 you would have to convert to 8bit before saving as a JPEG. In CS5 this conversion is handled automatically.

When editing in 16bit, Photoshop has more information to work with while you make adjustments. You will see more banding with certain adjustments when you edit in 8bit.

At the very end, Photoshop will convert your final product to 8bit. That change is usually barely noticable for most photographs.
