Best way to set up "micro adjustments" for a lens on a 50D

I think that the tamron on my 50D needs a slight "tweak" in terms of adjustment..

I guess the best way is to use a focus chart...solid tripod, remote release and perhaps the camera in tethered mode

Any other "hints" suggestions most welcome
