Good wildlife lens please?

Hi all,

Since a few years I'm really into photography. I currently own a Canon 20D and have various lenses. I use the 100-400 IS lens a my main wildlife lens but I would like to buy one of Canon's big guns :). I'll be waiting for a new body until a new Mark II is issued.

Can anyone give me advise on the lens to buy. I only want to buy a Canon lens , no third party. I'm thinking of the Canon 300 2.8 IS or the Canon 500 f/4 IS. Can they still be used hand held if needed? Quality wise there will be no difference I guess. And what about autofocus speed? Anyone used both and is able to give me some advise? The Canon 400 2.8 IS is even more expensive and bit above my planned budget. I can also imagine that it will be even more difficult using that one hand held occasionally.

I'm a big guy and sport a lot. My canon 20D including 100-400 lens I can use easily on a days hiking without any problems.
